In a large, community-based, open-label trial, 30 days of treatment with entacapone added to levodopa resulted in reduced frequency of levodopa administration and significant improvements in overall quality of life.43 Nearly 20% of patients reported depression at baseline, but that incidence was reduced by about 40% after just 30 days. Patients who were no longer depressed at study completion achieved the greatest improvement in quality-of-life scores.44 Entacapone is usually well tolerated when added to levodopa. Increased or prolonged dyskinesia and nausea can be managed by reducing the dose of levodopa. Entacapone also causes urine discoloration. Diarrhea has been reported with both entacapone and tolcapone. Entacapone is also marketed in a fixed combination that incorporates levodopa 50, 100, or 150 mg ; , carhidopa 12.5, 25, or 37.5 mg ; , and entacapone 200 mg.
